Abstract Protein-based biomaterials have the characteristics of stability and biocompatibility. Based on these advantages, various bionic materials been manufactured used in different fields. However, current protein-based generally need to form monomers cells be purified before being assembled vitro. The preparation process takes a long time, complex cellular environment is challenging optimized for producing target protein product. Here this study proposed technology situ synthesis assembly protein, namely cell-free (CFPS), which allowed shorten time increase flexibility adding or removing natural synthetic components. In study, successful expression self-assembly dihedral symmetric proteins proved applicability CFPS system production. Furthermore, fusion functional six scaffold could active polymers system.
